Die TUNEL-Methode (TdT-mediated dUTP-biotin nick end labeling) dient der Darstellung von Zellkernen apoptotischer Zellen. βTdTβ steht fΓΌr βterminal desoxynucleotidyl transferaseβ, ein Enzym, das in einem Zwischenschritt der Technik verwendet wird.

Methode
WΓ€hrend der Apoptose wird der DesoxyribonukleinsΓ€ure-Strang (DNA-Strang) des Zellkerns durch die AktivitΓ€t von Endonukleasen fragmentiert. Die an den Bruchenden freiwerdenden Hydroxygruppen (3'-OH-Gruppen) werden durch das Enzym TdT mit markierten Nukleotiden versehen, welche z. B. aufgrund von Fluoreszenz mit entsprechenden Mikroskopen sichtbar gemacht werden kΓΆnnen.
Die Methode wurde 1992 erstmals beschrieben.cite-ref-1[1] An der Methode wurde in den darauffolgenden Jahren Kritik geΓΌbt, da eine Unterscheidung zwischen nekrotischen und apoptotischen ZelluntergΓ€ngen nur bedingt mΓΆglich war.cite-ref-2[2] In den Jahren darauf wurde beschrieben, wie die Technik hinsichtlich auftretender Probleme modifiziert und verbessert werden kann.cite-ref-3[3]cite-ref-4[4]cite-ref-5[5]
